Well written, but morally objectionable
Incredibly arousing and well written(as always). Just one problem...
I have a moral objection to what the secretary is doing. She is attempting to drastically change her employer's entire life, without said employer's consent or even knowledge. That's going to lead to a lot of problems for her/him...
If this story has a romantic conclusion, I'd think it would involve elements of nonconsensual/BDSM as the new man of the office vents his lusts and his moral outrage at the same time...

An intriguing start. I can't wait to read your next installment. There are so many twists that you can create that I'm sure there will be some surprises!
It's just a fantasy
Since such a thing as sex-change via pills, does not exist then any moral objections don't matter. This is just a fantasy and whatever the author decides is acceptable goes. Keep writing.
moral objection
If the pill existed, Id have a moral objection to your story, but since it is truely fiction, should be an interesting series. One question, since you think she looks butch, how do you know she does not alreadt have a cock and does not hit on you because of sexual harresment.
I hope there are consequences
Its a good start, but I hope you don't make the boss weak. She should be outraged at her sex change, and demand it restored. If it can't be, she should then fire her secretary, and sue her for several million dollars for a forced, unwanted sex change. Relationships are built on trust, admiration, and respect. The secretary has shown she has none of these for the boss, and should suffer grieviously for what she does.
this story needs irony
Irony I:
this story cries out for Vivian, the butch looking boss, to be a secret (to brenda) pre-op male to female transexual.
then brenda's magic pill would not change Vivian to a male, like brenda wants, but just keeps Vivian a womanly woman.
Irony II:
Vivian changes to a man, loves it, becomes the most sexually obnoxious boss brenda ever had - but brenda is trapped working for 'him', since if she leaves she will go to jail.....

a mystery
I honestly think the story should end there. Not because of moral objections (as others have mentioned), but because I like the way it ended. Who needs the boss's reaction? The fantasy, and lack of conclusion, should be enough.
